Autoimmune Signaturen des Alterns und altersbedingter neurodegenerativer Erkrankungen stehen im Zusammenhang mit der Gehirnfunktion und ribosomalen  Proteinen

Die biologische Alterung ist ein komplexer Prozess, der durch eine verminderte Funktion von Zellen und Geweben, einschließlich derjenigen des Immunsystems, gekennzeichnet ist. Folglich beeinflusst das Altern die Expression und Entwicklung von Autoantikörpern und autoreaktiven T-Zellen, die in ihrer Summe als Autoimmunom eines Individuums gesehen werden können. In dieser Studie haben wir analysiert, ob Sätze von Autoimmunmerkmalen mit spezifischen Phänotypen assoziiert sind, die autoimmune Signaturen im Zusammenhang mit Alter und neurodegenerativen Erkrankungen bilden. Die Autoantikörperprofildaten von gesunden Probanden und Patienten aus der GEO-Datenbank wurden verwendet, um autoimmune Signaturen des Alterns und dreier neurodegenerativer Erkrankungen, darunter Parkinson-Krankheit (PD), Alzheimer-Krankheit (AD) und A number of Sklerose (MS), zu untersuchen.

Unsere Ergebnisse zeigen, dass die autoimmune Signatur des Alterns durch einen wellenförmigen Anstieg von IgG-Autoantikörpern im Zusammenhang mit Lernen und Verhalten und einem konsistenten Anstieg von IgG-Autoantikörpern im Zusammenhang mit Ribosomen und Translation gekennzeichnet ist Krankheiten. Interessanterweise identifizierte die Differential Expression-Sliding Window Evaluation (DE-SWAN) drei Wellen von Veränderungen von Autoantikörpern während des Alterns im Alter von 30, 50 bzw. 62 Jahren. Darüber hinaus sind insbesondere IgG-Autoantikörpersolche gegen ribosomale Proteine, könnten als Vorhersagemarker für das Altern und altersbedingte neurodegenerative Erkrankungen verwendet werden. Daher deckt diese Studie erstmals umfassende autoimmunologische Signaturen für das Altern und altersbedingte neurodegenerative Erkrankungen auf.

Kombination von schnellem mikrofluidischem Mischen und Dreifarben-Einzelmolekül-FRET zur Untersuchung der Kinetik von  Protein-  Konformationsänderungen

Der Einzelmolekül-Förster-Resonanzenergietransfer (FRET) ist aufgrund seiner hohen Empfindlichkeit und Fähigkeit, einzelne Subpopulationen in heterogenen Systemen aufzulösen, intestine geeignet, um die Kinetik von Protein-Konformationsänderungen zu untersuchen. Der gebräuchlichste Ansatz, der zwei Fluorophore verwendet, kann jedoch jeweils nur eine Distanz überwachen, und die Verwendung von drei Fluorophoren zum gleichzeitigen Überwachen mehrerer Distanzen ist weitgehend auf Gleichgewichtsschwankungen beschränkt. Hier zeigen wir, dass Dreifarben-Einzelmolekül- FRET mit schnellem mikrofluidischem Mischen kombiniert werden kann , um Konformationsänderungen in einem Protein von Millisekunden bis Minuten zu untersuchen. In Kombination mit manuellem Mischen haben wir die Kinetik auf 1 h verlängert, was einem Gesamtzeitbereich von 5 Größenordnungen entspricht.

Wir untersuchten die Umwandlung von Monomer zu Protomer des porenbildenden Toxins Cytolysin A (ClyA), einem der größten bekannten Konformationsübergänge von Proteinen. Die ortsspezifische Markierung von ClyA mit drei Fluorophoren ermöglichte es uns, die Kinetik von drei intramolekularen Abständen gleichzeitig zu verfolgen und zeigte ein bisher unentdecktes Intermediat. Die Kombination von dreifarbigem Einzelmolekül- FRET mit schneller mikrofluidischer Mischung bietet somit einen Ansatz, um die Mechanismen komplexer biomolekularer Prozesse mit hoher Zeitauflösung zu untersuchen.

Kollagene sind ein Hauptbestandteil der extrazellulären Matrix und sind funktionelle Liganden für den inhibitorischen Immunrezeptor- Leukozyten-assoziierten Immunglobulin-ähnlichen Rezeptor (LAIR)-1. LAIR-2 ist ein sezerniertes Protein, das als Köderrezeptor fungieren kann, indem es Kollagen mit einer höheren Affinität als LAIR-1 bindet. Wir schlagen vor, dass Kollagene die Immunevasion durch Interaktion mit LAIR-1 fördern, das auf Immunzellen exprimiert wird, und dass LAIR-2 eine LAIR-1-vermittelte Immunsuppression freisetzt.

Immunologische Identifizierung und Charakterisierung des Capsid Scaffold  Protein  kodiert durch UL26.5 von Herpes – simplex – Virus Typ 2

Herpes-simplex-Virus Typ 2 (HSV2), ein Erreger, der Genitalherpes-Läsionen verursacht, greift über  verschiedene bekannte und unbekannte Mechanismen in das Immunsystem des Wirts ein  . Dieses Virus wurde verwendet, um die virale Antigenzusammensetzung zu untersuchen. Rekonvaleszenzserum von HSV2-infizierten Patienten wurde verwendet, um virale Antigene mittels 2D-  Proteinelektrophorese und Immunblotting zu identifizieren  . Das Serum erkannte überwiegend mehrere Kapsidgerüstproteine, die vom Gen UL26.5, hauptsächlich ICP35, kodiert wurden .

Von diesem Protein wurde hauptsächlich berichtet, dass es vorübergehend beim viralen Zusammenbau funktioniert, aber nicht in reifen Viruspartikeln exprimiert wird. Weitere immunologische Studien legten nahe, dass dieses Protein bei Mäusen spezifische Antikörper- und zytotoxische T-Lymphozyten (CTL) -Reaktionen hervorruft, aber diese Reaktionen führen nicht zu einer klinischen Schutzwirkung als Reaktion auf eine HSV2-Provokation . Die Daten legten nahe, dass die Immundominanz von ICP35 verwendet werden könnte, um ein integriertes Antigen mit anderen viralen Glykoproteinen zu entwickeln.

 

Identifizierung von 7.000-9.000  Proteinen  aus Zelllinien und Geweben durch Single-Shot Microflow LC-MS/MS

Ein aktueller Pattern in der Proteomik besteht darin, Daten in einem „Single-Shot“ mit LC-MS/MS zu erfassen, da dies die Arbeitsabläufe vereinfacht und einen besseren Durchsatz und eine höhere quantitative Genauigkeit verspricht als Schemata, die eine umfangreiche Probenfraktionierung beinhalten. Single-Shot-Ansätze können jedoch unter einer begrenzten Proteomabdeckung leiden, wenn sie durch datenabhängige Erfassung (ssDDA) auf Nanoflow-LC-Systemen durchgeführt werden. Für Anwendungen, bei denen Probenmengen nicht knapp sind, zeigt diese Studie, dass eine hohe Proteomabdeckung mit einem Microflow-LC-MS/MS-System erreicht werden kann, das eine 1 mm ID × 150 mm Säule mit einer Flussrate von 50 μL/min und gekoppelt betrieben an ein Orbitrap HF-X-Massenspektrometer.

Die Ergebnisse zeigen die Identifizierung von 9 000 Proteinen aus 50 μg Proteinverdau aus  Arabidopsis-  Wurzeln, 7 500 aus Mausthymus und 7 300 aus menschlichen Brustkrebszellen in three h Analysezeit in einem einzigen Durchlauf. Der mit dem iBAQ-Ansatz gemessene dynamische Bereich der Proteinquantifizierung umfasste 5 Größenordnungen und die Wiederholungsanalyse zeigte, dass der mittlere Variationskoeffizient unter 20 % lag. Zusammen zeigt diese Studie, dass ssDDA durch μLC-MS/MS eine robuste Methode für umfassende und groß angelegte Proteomanalysen ist und in Zukunft auf schnellere Chromatographie- und datenunabhängige Akquisitionsansätze ausgeweitet werden kann.̀.
