Autoimmune Signaturen des Alterns und altersbedingter neurodegenerativer Erkrankungen stehen im Zusammenhang mit der Gehirnfunktion und ribosomalen Proteinen
Die biologische Alterung ist ein komplexer Prozess, der durch eine verminderte Funktion von Zellen und Geweben, einschließlich derjenigen des Immunsystems, gekennzeichnet ist. Folglich beeinflusst das Altern die Expression und Entwicklung von Autoantikörpern und autoreaktiven T-Zellen, die in ihrer Summe als Autoimmunom eines Individuums gesehen werden können. In dieser Studie haben wir analysiert, ob Sätze von Autoimmunmerkmalen mit spezifischen Phänotypen assoziiert sind, die autoimmune Signaturen im Zusammenhang mit Alter und neurodegenerativen Erkrankungen bilden. Unsere Ergebnisse zeigen, dass die autoimmune Signatur des Alterns durch einen wellenförmigen Anstieg von IgG-Autoantikörpern im Zusammenhang mit Lernen und Verhalten und einem konsistenten Anstieg von IgG-Autoantikörpern im Zusammenhang mit Ribosomen und Translation gekennzeichnet ist Krankheiten. Interessanterweise identifizierte die Differential Expression-Sliding Window Evaluation (DE-SWAN) drei Wellen von Veränderungen von Autoantikörpern während des Alterns im Alter von 30, 50 bzw. 62 Jahren. Der Einzelmolekül-Förster-Resonanzenergietransfer (FRET) ist aufgrund seiner hohen Empfindlichkeit und Fähigkeit, einzelne Subpopulationen in heterogenen Systemen aufzulösen, intestine geeignet, um die Kinetik von Protein-Konformationsänderungen zu untersuchen. Der gebräuchlichste Ansatz, der zwei Fluorophore verwendet, kann jedoch jeweils nur eine Distanz überwachen, und die Verwendung von drei Fluorophoren zum gleichzeitigen Überwachen mehrerer Distanzen ist weitgehend auf Gleichgewichtsschwankungen beschränkt. Hier zeigen wir, dass Dreifarben-Einzelmolekül- FRET mit schnellem mikrofluidischem Mischen kombiniert werden kann , um Konformationsänderungen in einem Protein von Millisekunden bis Minuten zu untersuchen. In Kombination mit manuellem Mischen haben wir die Kinetik auf 1 h verlängert, was einem Gesamtzeitbereich von 5 Größenordnungen entspricht.
Wir untersuchten die Umwandlung von Monomer zu Protomer des porenbildenden Toxins Cytolysin A (ClyA), einem der größten bekannten Konformationsübergänge von Proteinen. Die ortsspezifische Markierung von ClyA mit drei Fluorophoren ermöglichte es uns, die Kinetik von drei intramolekularen Abständen gleichzeitig zu verfolgen und zeigte ein bisher unentdecktes Intermediat. Die Kombination von dreifarbigem Einzelmolekül- FRET mit schneller mikrofluidischer Mischung bietet somit einen Ansatz, um die Mechanismen komplexer biomolekularer Prozesse mit hoher Zeitauflösung zu untersuchen.
